Prof. Brenda Wilson from the University of Illinois at Urbana-Champaign will present the following talk on bacterial toxins: 

Title: G-Protein-Deamidating Bacterial Protein Toxins as Modulators of Zoonotic Disease and Cancer Risk

Abstract: AB-type bacterial protein toxins are being explored as cytosolic delivery vehicles in biomedical and research applications due to their modular nature, potency, and cell specificity. Full-length toxin homologs of the cytotoxic necrotizing factors (CNFs) are found in a wide array of bacterial species, including insect, fish, livestock, and human pathogens. CNF intoxication results in changes in cell morphology, motility, gene expression, cell survival, oncogenic transformation, tumorigenesis and metastasis, hallmarks of cancer risk. These toxins contain an N-terminal delivery vehicle, which consists of the functional domains for receptor binding and translocation, and a C-terminal cargo domain that, upon translocation from the acidifying endosome, is cleaved from the rest of the toxin and delivered into the host cell cytosol, where they catalyze irreversible deamidation of their target Rho-G-protein substrates. Utilizing the cargo delivery systems of the CNF family, we have identified those domains that exhibit a universal ability to deliver or be delivered efficiently into the cell cytosol. Further study of how these highly homologous domains have co-evolved to accommodate one another may identify determinant regions that dictate compatibility of the cargo with its delivery vehicle to enable efficient cytosolic delivery.
